I arrived in Venice on May 28, 2005 and stayed for five days. The first thing I did was buy a vaparetto pass (well worth the cost). Dinner is expensive in this city so plan on spending at least $40 per night if you go to a nice/decent restaurant. I also bought a choir pass to the churches (good value). The weather was wonderful. I spent my time just walking the narrow streets of Venice, ducking into churches or museums or riding on the vaparetto (work your way to the front or back). The canals were clean and no mosquitoes.
I went to the islands of Murano (glass) and Burano (lace), but unless you are really into watching glass made-don't bother. Also Burano WAS the place for lace, but no more. The lace is imported from China and here is the only place I saw dirty canals. Lido is about a 20 minute vaparetto ride across the lagoon to the Doge's place and a good alternate if you do not want to stay directly in Venice.
One day there was a strike by the vaparetto drivers. Venice is small and easy to walk around. I did see many tourists with huge roller bags struggling to climb up/down the many little bridges and narrow walkways.
One last thing, if you ask for directions, you will be told at some point to go "straight" down a street. Straight in Venice is not the same anywhere else. Invest in a good map. Also, the way Venetians number addresses is something no one outside Venice will ever figure out.
I am a single female who traveled alone to Venice and felt safe walking around the central area. Venice is a beautiful and unique city and I look forward to a return visit.
