If you want to fly to get to Elko, then you can fly to Elko Regional Airport , which is located only one mile from downtown Elko. This is a very small airport, and there are actually only three runways for the entire area. There is only one commercial airline that flies in-- Skywest Airlines . There are some other companies that fly in as well, but they aren't as large as Skywest Airlines. You can also get private chartered flights to the airport. Once you get into Elko you can get a taxi to take you to where you want to go. You can also get a rental car from the airport if you want to drive around town when you get in. You don't just have to fly to get into Elko though. You can also take a train or a bus. Buses and trains make several trips to the town every day. There are different times which are posted at the stations. It usually isn't too hard to get tickets, but you should plan ahead just to be safe. If you want to, you can drive to Elko also. The traffic should not be too bad to get into town, and it's a good way to save money on taxis, rental cars, and plane/train/bus tickets.
UPDATE: DO NOT take a bus to Elko. Locals call it ConGreyhound, so unless you want to make some unsavory connections and be deposited in a desolated place with no building in the pitch black night, don't bother with this. It's not just a bad idea, it's not safe. As for the train, love it but it will be late. Always is, always will be. SkyWest, booked through Delta, does a wonderful job of supplying service to small towns west of Colorado. The hub is in Salt Lake City and from there it's a 60-minute flight to Elko several times a day.
